Use screws for the main structure. You can use nails for some support structures. Screws are easier if your not that skilled with a hammer, it just takes longer. If you cladding then you will want to use nails.

Screwfix all the way for screws/nails. Loads cheaper than DIY stores.

Depends. Galvanized nuts and bolts or lag bolts for most heavy beam and timber, nails for floor joist hangers and wall studs, and galvanized nails and screws for exterior siding. I also used construction adhesive in critical areas, including subfloors so I don't get the squeaks.
