Vignette, alignment and much puzzlement

Was watching a vid the other day about vignetting and flat frames so just had a play... the top part of the image below is the initial pic i took and ran through CCDInspectors flat frame analysis and the bottom part is the final pic after tweaking the optical train in my scope.

The problem now is that  laser collimator says the collimation is a mile off. 

Any experts tell me what's going on? because i'd have thought that centering the vignette would show that the optical train is collimated with the dslr
