I find it best to leave them as they are. FYI The gain is like turning up the volume control, it is how much the amplifier in the DSI erm.. amplifies the signal. It's basically like the ISO control on a DSLR.

The Offset is, a small amount that is added to the pixels so that they always register something. That's not a very good explanation and I cannot think of an analogy so.... Each pixel in the CCD array gets a digital number assinged to it based on the charge gathered over the exposure, this is what gives each pixel a diffrent brightness. The offset is added to the digital number so that it never reads zero. I think!!! I also dont know why this is important either!
