I have a Nexstar 4se and canon EOS 500D Mark III and want to know the Step by step from turning the scope and camera on to taking a good photo of andromeda.

Unfortunately your scope has a very long focal length so you won’t be able to fit the whole of the Andromeda Galaxy in your camera frame. Have a look at this website to give you an idea of the field of view of different telescope and camera combinations:

Have you tried photographing the moon or planets with your scope? They would be better targets to start with.
