Have you tried the wavelets in registax?

Also maybe the focus is slightly off?

And finally you might get more feedback in the imaging section, not visual.

Something is wrong. Plausably the "contrast" or other setting is to high (a lot). You don't sharpen the image by camera settings, but later when processing the final stack... so keep those camera settings default as much as possible (except gain and exposure time)
